Docking protein 7 (DOK7) is an intracellular adaptor protein primarily involved in signal transduction. It is essential for activating muscle-specific kinase (MuSK), which is required for the formation and maintenance of the neuromuscular junction. Mutations in DOK7 are known to cause a form of congenital myasthenic syndrome, leading to neuromuscular transmission defects. In addition to its role in muscle biology, DOK7 has been implicated in cancer biology, known to suppress the progression of certain cancers, such as bladder cancer, where it is a target of microRNAs like miR-299-5p. There is limited evidence of DOK7 being directly modulated by small-molecule drugs or being targeted for therapeutic intervention, but its critical physiological roles make it a subject of ongoing research.
