Dolichyl-phosphate N-acetylglucosaminephosphotransferase 1 (DPAGT1) is an essential enzyme in the process of protein N-glycosylation. It catalyzes the first committed step in the dolichol-linked oligosaccharide pathway, which is crucial for glycoprotein biosynthesis in eukaryotic cells. It transfers N-acetylglucosamine (GlcNAc) from uridine diphosphate N-acetylglucosamine (UDP-GlcNAc) to dolichyl-phosphate, forming dolichyl-phosphate N-acetylglucosamine and UMP. Mutations or dysfunctions in DPAGT1 can result in congenital disorders of glycosylation (CDG), affecting multiple organ systems due to defective protein glycosylation. The best-known inhibitor is tunicamycin, which blocks eukaryotic protein N-glycosylation.
