Donor immune cells from mobilized peripheral blood are a collection of immune cells, including hematopoietic stem and progenitor cells, lymphocytes, monocytes, and dendritic cells, obtained from a donor after stimulation with agents like G-CSF or plerixafor. This process mobilizes cells from the bone marrow into the bloodstream, allowing for collection via leukapheresis. These mobilized cells are used primarily for hematopoietic stem cell transplantation and cellular therapies, offering a higher yield of stem cells compared to steady-state collections. G-CSF mobilization can induce immunosuppressive effects, shifting the balance towards regulatory T-cells with altered NK cell activity. Quality control measures involve screening for infectious diseases and ensuring ethical sourcing.
Mobilization agents (G-CSF, Plerixafor, Motixafortide) disrupt the interaction between hematopoietic stem and progenitor cells and the bone marrow microenvironment, specifically by interfering with the CXCR4/CXCL12 axis, leading to increased release of these cells into the peripheral blood. The mobilized cells are then collected via leukapheresis and used for transplantation or cellular therapy.
