The Dopamine D1-like receptor family (comprising Dopamine receptor D1 and Dopamine receptor D5) represents a group of G protein-coupled receptors that activate adenylyl cyclase via Gs and Golf protein coupling, increasing intracellular cAMP and modulating neural activity. D1-like receptors are primarily located in the striatum, cortex, limbic regions, and other areas of the central nervous system. They regulate motor control, cognition, reward, and emotion and are pivotal in both normal physiology and pathological conditions such as Parkinson’s disease, schizophrenia, and addiction. Pharmacological agents targeting these receptors include agonists, antagonists, and indirect modulators, with ongoing therapeutic research in neuropsychiatric and neurodegenerative disease settings. The D1 receptor especially is the most abundant dopamine receptor in the brain and displays a distinct regulation of gene expression and neuronal function within dopaminergic circuits.
