Overview

The dopamine D1 receptor (D1R) is a member of the G protein-coupled receptor (GPCR) family, specifically classified within the D1-like subfamily. It is the most abundant dopamine receptor subtype in the central nervous system and plays a key role in regulating motor behavior, reward processing, motivation, memory formation, learning, attention, impulse control, and neuronal development. Dysfunctional signaling is associated with neuropsychiatric disorders such as Parkinson’s disease (PD), schizophrenia, Huntington's disease, attention deficit hyperactivity disorder (ADHD), cognitive impairment disorders—and plays a role in addiction mechanisms. The D1R is an established therapeutic target for CNS diseases including PD.

Mechanism of action

Activation stimulates adenylyl cyclase via Gαs/olf coupling, increasing intracellular cAMP levels and activating protein kinase A (PKA).
