Overview

The Dopamine D3 receptor is a protein encoded by the DRD3 gene. It is a D3 subtype of dopamine receptor and functions as a G protein-coupled receptor, inhibiting adenylyl cyclase via inhibitory G-proteins. It is primarily expressed in phylogenetically older brain regions like the ventral striatum (nucleus accumbens) and other limbic areas, with lower levels in the dorsal striatum and cortex. The receptor functions both as an autoreceptor and a post-synaptic receptor and plays crucial roles in attention, memory, emotions, motivation, reward, cognitive functions, emotional regulation, and promoting neuroprotection. Due to its specific distribution, it is a significant therapeutic target for conditions like schizophrenia (especially negative symptoms), drug addiction, Parkinson's disease, and depression, offering the potential to develop drugs with fewer motor side effects compared to D2 receptor antagonists. The close homology with the D2 receptor presents challenges in developing highly selective compounds.
