Overview

The dopamine receptor D1 (DRD1) is a G protein-coupled receptor and a member of the dopamine receptor family. It is the most abundant dopamine receptor in the central nervous system and plays key roles in reward processing, motor control, and cognitive functions. DRD1 primarily couples to Gαs/olf, leading to stimulation of adenylyl cyclase and increased cAMP levels. Dysfunctional DRD1 signaling has been implicated in neuropsychiatric disorders such as Parkinson’s disease, schizophrenia, and ADHD. Current pharmacotherapies targeting this pathway focus on PD and cognitive impairment but also play roles indirectly via drugs like L-DOPA.
