Overview

The D2-like dopamine receptor family comprises D2, D3, and D4 receptor subtypes, all of which are G protein-coupled receptors that bind dopamine. These receptors are primarily Gi/o-coupled, leading to inhibition of adenylyl cyclase activity. They play crucial roles in regulating dopamine release, neuronal excitability, and various behavioral functions, including locomotion, attention, and cognition. The D2 receptor is a primary target for antipsychotic drugs, and alterations in D2-like receptor function have been implicated in neuropsychiatric disorders such as schizophrenia, Parkinson's disease, and ADHD.
