Overview

The dopamine receptor D3 (DRD3) is a G protein-coupled receptor primarily expressed in the ventral striatum, thalamus, hippocampus, and cortex. It modulates dopamine synthesis and release, regulates cognition, emotion, and motivation, and is implicated in schizophrenia, drug addiction, Parkinson's disease, and depression. It is a target for antipsychotics and drugs used to treat Parkinson's disease and addiction.

Mechanism of action

Inhibition of adenylyl cyclase via Gi/o proteins upon dopamine binding. Acts as an autoreceptor on dopaminergic neurons to modulate dopamine synthesis and release; also present postsynaptically where it influences neuronal signaling pathways.
