The dopamine transporter (DAT), encoded by the SLC6A3 gene, is an integral membrane protein responsible for clearing dopamine from the synaptic cleft by transporting it back into presynaptic neurons. This reuptake process regulates extracellular dopamine levels, which are critical for various aspects of cognition, including reward processing. DAT function is implicated in several neuropsychiatric disorders and is the direct target of psychostimulants like cocaine and amphetamines.
Cocaine blocks reuptake of dopamine by DAT; amphetamines reverse the transport direction of DAT, leading to increased synaptic dopamine concentrations.
