Overview

Dopaminergic neuron differentiation is the process by which precursor or stem cells develop into mature dopaminergic neurons—neurons that synthesize and release dopamine as their primary neurotransmitter. This process is critical for both normal brain development and for regenerative medicine approaches, particularly in the context of neurodegenerative diseases such as Parkinson’s disease. Key stages include neural induction, midbrain floor plate specification, progenitor expansion, and maturation. It is regulated by multiple signaling pathways (SMAD/BMP/TGFβ, SHH, Wnt, Notch) and defined by expression of stage-specific markers (OTX2, EN1, FOXA2, LMX1A, NEUROG2, TH, PITX3).
