The Dorsolateral Prefrontal Cortex (dlPFC) neuronal circuits are essential components of the primate brain responsible for executive functions, including working memory, cognitive flexibility, and planning (StatPearls, 2023). These circuits consist of interconnected pyramidal neurons in layer III that excite one another to maintain persistent firing, a process regulated by GABAergic interneurons and modulated by catecholamines like dopamine and norepinephrine (Arnsten, 2011). Impairment of dlPFC circuitry is strongly linked to the cognitive deficits observed in schizophrenia, ADHD, and major depressive disorder, where the tuning of these neurons is disrupted (Goldman-Rakic, 1995). While not a single molecular target, the dlPFC is the primary site for neuromodulation therapies and a key region for drugs targeting alpha-2A adrenoceptors or D1 dopamine receptors to improve cognitive signal-to-noise ratios. Understanding these circuits is vital for developing circuit-based therapeutics that aim to restore functional connectivity in the brain (National Institute of Mental Health, 2023).
