Double C2-like domain-containing protein alpha (DOC2A) is a calcium-sensing, tandem C2-domain protein that binds both calcium and phospholipids, likely regulating fusion of vesicles with membranes and facilitating calcium-dependent neurotransmitter release in neuronal cells. DOC2A is predominantly expressed in the brain and may also regulate secretory lysosome exocytosis in mast cells. It interacts with proteins such as UNC13A and UNC13B, and its dysfunction is associated with neurological phenotypes—particularly neurodevelopmental disorders related to chromosome 16p11.2. While essential in synaptic vesicle exocytosis, it is not recognized as a direct therapeutic target, and there are no clinically relevant drugs known to target DOC2A directly.
