Double C2-like domain-containing protein beta (DOC2B) is a tandem C2-domain calcium sensor protein involved in calcium-dependent vesicle trafficking and exocytosis. It is expressed in neuronal tissues, where it regulates spontaneous and asynchronous neurotransmitter release, as well as in endocrine cells such as pancreatic β-cells, where it is critical for glucose-stimulated insulin secretion. DOC2B acts by binding calcium and phospholipids, facilitating SNARE-dependent membrane fusion. Structurally, its C2B domain is the primary high-affinity calcium sensor, while the C2A domain assists in membrane interaction. Decreased DOC2B expression is associated with impaired insulin release, and its secretion in extracellular vesicles may reflect β-cell functional status. DOC2B is not a known pharmacological target, but it plays essential roles in neuroendocrine secretion physiology
