Double homeobox A pseudogene 1 (DUXAP1)

Target
DUXAP1
Molecular classification
Other, Long non-coding RNA (lncRNA) pseudogene
01

Overview

Double homeobox A pseudogene 1 (DUXAP1) is a long non-coding RNA pseudogene implicated as an oncogenic driver in multiple malignancies, with key roles in promoting cell proliferation, migration, invasion, and chemotherapy resistance. In hepatocellular carcinoma specifically, DUXAP1 acts as a ceRNA by sequestering miR-584-5p, thereby upregulating MAPK1 (ERK2) and activating the MAPK/ERK pathway, which contributes to increased malignancy and drug resistance. DUXAP1 expression is upregulated via m6A methylation and is positively associated with tumor progression, recurrence, and poor clinical outcomes.

Mechanism of action

DUXAP1 acts as a molecular sponge for miR-584-5p via ceRNA mechanism, upregulating MAPK1 (ERK2) and activating MAPK/ERK signaling, thereby promoting malignant phenotype and drug resistance.

Biological functions

Regulation of cell proliferationPromotion of cell migration and invasionEnhancement of drug resistance (specifically chemotherapy resistance in hepatocellular carcinoma)Regulation of cancer stemness pathwaysOncogenic ceRNA (competitive endogenous RNA) regulatory function
04

Disease associations

Cancer (especially hepatocellular carcinoma, HCC)Therapy resistance (chemotherapy resistance)Tumor growth and metastasis (e.g., increased migration, invasion, and stemness markers in malignancy)
05

Safety considerations

Targeting lncRNAs such as DUXAP1 poses challenges including off-target effects, lack of protein knockdown specificity, and potential systemic toxicity due to regulatory network complexity
06

Interacting drugs

Sorafenib
07

Biomarkers

DUXAP1 (expression is correlated with HCC progression, metastasis, and chemoresistance)MAPK1 (as downstream effector of DUXAP1-driven pathway)Stemness genes: CD133, OCT4, NANOG, Sox2 (their expression is influenced by DUXAP1 activity)
