Double homeobox A pseudogene 9 (DUXAP9) is a long non-coding RNA (lncRNA) transcribed from a pseudogene related to the double homeobox gene family. Although originally classified as a pseudogene, DUXAP9 is transcribed and acts as a regulatory lncRNA in the nucleus. It physically interacts with the histone methyltransferase EZH2, stabilizing EZH2 by inhibiting its phosphorylation and subsequent degradation, which in turn promotes cell proliferation, migration, invasion, and epithelial–mesenchymal transition—especially in tumors such as oral squamous cell carcinoma and clear cell renal cell carcinoma[1][2][3][4]. DUXAP9 is associated with poor prognosis in multiple cancer types and has been proposed as a prognostic biomarker and potential therapeutic target. No drugs currently target DUXAP9 directly, but it functions as an important modulator of epigenetic machinery and cancer pathways[2][4].
