Double homeobox protein 4-like protein 3 (DUX4L3) is a human pseudogene in the homeobox gene family[7][8]. It is highly homologous to the functional DUX4 gene, but it does not have evidence of protein-coding function. DUX4L3 is located within a macrosatellite repeat array on chromosome 4, an arrangement typical for certain homeobox pseudogenes. There is no evidence that DUX4L3 acts as a functional transcription factor as DUX4 does or that it is involved in disease beyond potential sequence similarity and confusion with DUX4[7][8]. Disease roles, molecular functions, and therapeutic relevance pertain to DUX4, not DUX4L3. Key caution: DUX4L3 should not be confused with DUX4, which is a functional transcription factor with roles in embryogenesis and certain muscle diseases[1][3]. DUX4L3 itself is primarily a sequence placeholder without demonstrated protein product or functional impact in humans[7][8].
