Double homeobox protein B (DUXB)

Target
DUXB
Molecular classification
Transcription factor, Homeobox protein, PRD-like homeobox family
01

Overview

Double homeobox protein B (DUXB) is a PRD-like homeobox transcription factor encoded by the human DUXB gene. The DUX family contains several homologous genes with roles in embryonic gene regulation and chromatin remodeling; DUXB has two homeodomains and is predicted to bind DNA and regulate transcription by RNA polymerase II. DUXB is distinct from the related DUX4, which is pathogenic in facioscapulohumeral muscular dystrophy (FSHD); DUXB itself is transcriptionally inactive in most tested assays and is considered to have limited or no transcriptional activation capacity, with unclear biological function and disease relevance. Alternative meanings: There is some ambiguity as "double homeobox" may refer to other DUX family proteins (like DUXA, DUX4), but "DUXB" specifically refers to Double homeobox protein B in humans. No major errors or ambiguities with this target designation were identified.

Disease associations

Potential involvement in Facioscapulohumeral muscular dystrophy (FSHD) types 1 and 2, digenic (disease association not well-defined, sometimes listed due to similarity with paralog DUX4)Other (the role is uncertain; some DUX family members impact embryogenesis and cancer, but DUXB itself has not been robustly linked to major diseases)
