Overview

RAD21L1 encodes a meiosis-specific cohesin complex subunit of the α-kleisin family, highly expressed in reproductive tissues such as testis and ovary[2][4]. The protein localizes to chromosome axes during the leptotene and zygotene stages of meiosis and is critical for synapsis and separation of homologous chromosomes[1][2][6]. In vertebrates such as zebrafish and mice, loss of RAD21L1 impairs oogenesis, causing defects in chromosome synapsis and triggering checkpoint-mediated cell death, leading to poor gamete quality and reduced fertility, while spermatogenesis is less affected[1]. RAD21L1 is essential for meiotic chromosome structure and integrity, but it is not currently a direct clinical or pharmacological target[1][6].
