Double-stranded RNA-dependent protein kinase (PKR) is a serine/threonine kinase that plays a central role in the cellular response to various stress signals, particularly viral infection. It is activated primarily by double-stranded RNA (dsRNA), leading to autophosphorylation and activation of downstream signaling pathways, including inhibition of translation, induction of apoptosis, and activation of inflammatory responses. Dysregulation or excessive activation of PKR can contribute to various pathologies, including autoimmune disorders, neurodegenerative diseases, cancer, and metabolic disorders.
