Double zinc ribbon and ankyrin repeat-containing protein 1 (DZANK1) is a structural protein containing two ankyrin repeat-encoding regions and zinc ribbon domains, which facilitate protein-protein interactions[1][2][4]. DZANK1 is involved in intracellular dynein-based transport, particularly essential for the vesicular trafficking required in photoreceptor cells of the retina[1][2][5][6][7]. Its primary function is linked to the assembly and folding of the cytoplasmic dynein 1 motor complex, influencing photoreceptor outer segment formation and organelle positioning. DZANK1 interacts with complementary subunits of dynein 1, specifically the light chains DYNLL1 and DYNLL2, supporting vesicle transport in cilia and photoreceptors[1]. Deficiency in DZANK1 causes specific retinal phenotypes in animal models, highlighting its contribution to ciliopathies and possible involvement in certain neurodegenerative or ophthalmic disorders. There is no documented evidence of drugs directly targeting DZANK1 nor established biomarker or safety profiles.
