Doublesex- and mab-3-related transcription factor A1 (DMRTA1) is a member of the DM-domain transcription factor family, characterized by its sequence-specific DNA-binding activity[1][5]. DMRTA1 plays a multifaceted role in neural development and cell fate determination, particularly in the mammalian forebrain, where it is activated in embryonic cortical progenitors by Pax6 to promote neurogenesis and neuronal lineage specification[1]. It also regulates sex differentiation and is predicted to act upstream of or within pathways related to male mating behavior and ovarian follicle development[1][5]. DMRTA1 localizes to chromatin and is predominantly nuclear[1][5]. Variations in the DMRTA1 locus have been associated with tumorigenic processes, particularly in IDH wild-type lower-grade gliomas, where precise gene dosage is necessary for normal cortical differentiation and genomic stability[1]. While DMRTA1 itself is not currently known to be a therapeutic drug target, its critical roles in neurodevelopment and association with disease make it biologically significant[1][5].
