Doublesex- and mab-3-related transcription factor A2 (DMRTA2) is a transcription factor within the DMRT family, characterized by a DM DNA-binding domain that is conserved from Drosophila (doublesex) and Caenorhabditis elegans (mab-3), both involved in developmental regulation[1][2][3]. DMRTA2 plays a critical role in the developing central nervous system, particularly by regulating the balance between proliferation and differentiation of neural progenitor cells (NPCs) during cortical development: it maintains NPCs in the cell cycle and inhibits their premature differentiation into neurons through direct transcriptional control of genes like Hes1[1][2]. Loss of function can cause cortical developmental disorders such as microcephaly. There is also evidence for possible involvement in sexual development, although this role is less defined compared to its neurodevelopmental functions[3]. DMRTA2 is not known to be a current therapeutic target, there are no established interacting drugs, and it is not used as a biomarker in clinical settings.
