Overview

DMRTB1 is a nuclear transcription factor that contains a conserved DM domain and belongs to the DMRT family. It plays a critical role in regulating the transition from mitotic spermatogonia to meiotic spermatocytes, marking a subset of germ cells in developing testes. Changes in its dosage or expression, such as those caused by genetic microdeletion, are strongly associated with impaired spermatogenesis and can lead to male infertility. DMRTB1’s expression is highly tissue-specific—primarily in testis—and its normal function is essential for male germ cell differentiation. Comparative genomics suggests that changes in DMRTB1 contribute to testis size evolution across tetrapods. Although principally involved in reproductive tissue development, recent genetic studies indicate potential additional systemic roles, including influence over gut microbial populations.
