Overview

Doublesex and mab-3 related transcription factor 2 (DMRT2) is a member of the DMRT family of transcription factors characterized by the DM DNA-binding domain, which is highly conserved and relates to sex determination genes in insects and nematodes[4][3]. DMRT2 is expressed predominantly during embryonic development, playing key roles in somitogenesis, neural patterning, and especially in endochondral bone formation by regulating chondrocyte maturation[2][5][1]. DMRT2 is regulated by upstream factors such as Sox9 and interacts with Runx2 in chondrocytes, and its loss leads to delayed bone formation and developmental abnormalities[2]. In the nervous system, it is implicated in region-specific neurogenesis and the specification of neural progenitors, independently of the Sonic hedgehog pathway[1][5]. DMRT2 is located on human chromosome 9p24.3, a region associated with disorders of sex differentiation, although its direct role in human gonadal development remains less clear[4]. There are currently no known drugs that modulate DMRT2 function, and no approved clinical biomarkers or therapies utilize this target.
