Overview

Doublesex and mab-3 related transcription factor 3 (DMRT3) is a member of the DMRT family of transcription factors, characterized by a conserved DM DNA-binding domain. It plays critical roles in sexual differentiation, neural development (particularly in spinal cord and telencephalon patterning), and has been implicated in tumorigenesis. In vertebrates, DMRT3 is essential for the specification of spinal interneurons that coordinate locomotor networks, as shown in mice and horses. Aberrant expression of DMRT3 is associated with poor prognosis in several human cancers and may influence cancer progression by modulating pathways such as cell cycle, apoptosis, EMT, and DNA repair. Mutations in DMRT3 underlie altered locomotion patterns in horses and may be involved in human disorders of sex development. While DMRT3 is associated with drug sensitivity and resistance in cancer cell lines, it is not yet a direct target of approved therapeutics, and its role as a biomarker and potential therapeutic target is under active investigation
