Dovitinib is an orally active, investigational small-molecule inhibitor of multiple receptor tyrosine kinases (including FGFR, VEGFR, PDGFR, KIT, and FLT3) that are involved in tumor growth, proliferation, survival, and angiogenesis. It is tested (but not FDA-approved) for the treatment of several cancers, including renal cell carcinoma, breast cancer, and melanoma, primarily as a third-line therapy or in combination with other drugs. Dovitinib works by inhibiting phosphorylation of these RTKs, reducing cellular proliferation and inducing apoptosis and autophagic cell death in tumor cells. Dovitinib is a benzimidazole-quinolinone compound. It should not be confused with a molecular target, as it is a therapeutic agent designed to block several kinases implicated in cancer pathogenesis.
