Overview

Dpy-30 histone methyltransferase complex regulatory subunit (DPY30) is an evolutionarily conserved core component of the SET1/MLL (KMT2) family of histone H3K4 methyltransferase complexes, also called COMPASS and COMPASS-like complexes[4][5][1]. DPY30 regulates global levels of histone H3 lysine-4 trimethylation (H3K4me3), a crucial epigenetic mark associated with transcriptional activation[4][2]. It stimulates the catalytic activity and processivity of MLL/SET1 complexes, especially on nucleosomal substrates rather than free histones, by forming a multiprotein module with ASH2L and RbBP5[2]. DPY30’s interaction with the intrinsically disordered regions of ASH2L is essential for establishing high levels of H3K4me3 and loci-specific histone methylation, modulating chromatin structure and gene expression during cellular differentiation, especially in hematopoietic and embryonic stem cells[2][4][5][8]. Loss or disruption of DPY30 results in genome-wide defects in H3K4me3 levels and thus impairs processes such as cell differentiation, proliferation, and lineage commitment, contributing to the development of cancer and other epigenetic diseases[4][8]. Despite its fundamental biological functions and role in disease, there are currently no approved drugs directly targeting DPY30, nor established clinical biomarkers or known unique safety concerns specific to modulation of DPY30 at this time.
