DPYD antisense RNA 1 (DPYD-AS1) is a long non-coding RNA (lncRNA) transcribed from the DPYD locus on chromosome 1p21.3.[2][3][5] It belongs to the family of natural antisense transcripts (NATs) and lies on the forward DNA strand at the DPYD locus, overlapping with the DPYD gene, which encodes dihydropyrimidine dehydrogenase.[2] DPYD-AS1 itself does not code for a protein but is believed to regulate the expression of the DPYD gene through epigenetic mechanisms, potentially impacting the breakdown of pyrimidines and drug response, especially regarding chemotherapeutics like 5-fluorouracil.[2][3] Variants or dysregulation of DPYD-AS1 may influence susceptibility to dihydropyrimidine dehydrogenase deficiency and associated adverse drug reactions, notably in cancer chemotherapy, by altering DPYD gene expression. There is currently no evidence that DPYD-AS1 is a direct pharmacological or therapeutic target, nor are there drugs known to interact with this RNA transcript directly.[2][3] Key points: - DPYD-AS1 is not a receptor, enzyme, or classical drug target but a regulatory lncRNA. - It may play a role in disease by modulating expression of the adjacent protein-coding gene DPYD, which is extremely significant for drug metabolism and toxicity, especially in cancer chemotherapy. - No direct drugs, mechanisms of action, or biomarker use described for DPYD-AS1 itself. - Research on its biological and disease roles is growing and mainly centers on regulation of gene expression and epigenetic modulation.[2][3]
