Overview

Dr1-associated corepressor (DRAP1) is a nuclear protein encoded by the DRAP1 gene, functioning as a corepressor of transcription. DRAP1 forms a complex with DR1, interacting with the TATA-binding protein (TBP) in eukaryotes. This complex selectively represses transcription initiation by preventing the entry of essential transcription factors (TFIIA, TFIIB) into the preinitiation complex, thereby regulating gene expression. This interaction is critical for stabilizing TBP-DR1-DNA complexes and modulating RNA polymerase II and III activities. Both DR1 and DRAP1 contain histone-fold motifs essential for their interaction and repression function. DRAP1 plays a central role in global transcriptional repression and cell viability, with dysfunction implicated in various diseases including some cancers
