Overview

DSN1 is a structural subunit of the MIS12 kinetochore complex, a core component of the outer kinetochore. The MIS12 complex (MIS12C), which includes DSN1, NSL1, MIS12, and PMF1, acts as a critical bridge linking the inner centromere (CCAN via CENP-C) with the outer kinetochore microtubule-binding sites (NDC80 complex). DSN1 is essential for proper chromosome alignment and segregation during mitosis. Aurora B kinase regulates the MIS12 complex function by phosphorylating DSN1, facilitating kinetochore assembly and tension-sensitive error correction. Disruption of DSN1 impairs kinetochore formation, chromosome segregation, and cell division, leading to chromosomal instability—a hallmark of many cancers[1][2][3][5][6]. DSN1 is not a direct therapeutic target, receptor, or enzyme, but its role in mitosis makes it important for understanding mechanisms of chromosomal instability in disease contexts.
