Overview

Dual specificity phosphatase 21 (DUSP21) is a member of the low molecular weight dual specificity phosphatase family, functioning as a protein tyrosine/serine/threonine phosphatase that localizes to both the cytoplasm and nucleus[1][7]. It is capable of removing phosphate groups from phosphotyrosine and phosphothreonine residues of substrate proteins, with an enzymatic preference for phosphotyrosine and diphosphorylated forms over phosphothreonine[1][6]. DUSP21 is implicated in the regulation of sperm motility by mediating dephosphorylation of sperm microtubule proteins, though its precise physiological substrates beyond synthetic MAPK peptides remain to be fully elucidated[1][6]. Clinically, variations in DUSP21 have been linked to neurodevelopmental disorders such as intellectual developmental disorder X-linked 109 and autism spectrum disorder[1]. No approved drugs or experimental compounds directly targeting DUSP21 were identified in available literature and database summaries. No established roles for DUSP21 as a therapeutic biomarker, nor any documented specific mechanism of action of drugs, are currently known. DUSP21 is considered an enzyme target, but there is currently limited evidence regarding its role in disease outside genetic association; as such, its druggability and therapeutic relevance remain to be established[1][6].
