Dual specificity phosphatase 28 (DUSP28)

Target
DUSP28
Molecular classification
Enzyme, Protein tyrosine phosphatase, Dual-specificity phosphatase, Atypical DUSP subfamily
01

Overview

Dual specificity phosphatase 28 is an enzyme belonging to the atypical DUSP subfamily. It catalyzes the removal of phosphate groups from tyrosine and threonine residues of protein substrates, but has atypically low intrinsic catalytic activity due to steric hindrance in its active site. DUSP28 is implicated in cell signaling regulation, particularly as a modulator of the MAPK/ERK pathway, and is associated with the progression, migration, and drug resistance of pancreatic and hepatocellular carcinomas. It is being investigated as a potential biomarker and therapeutic target in oncology

Other names
DUSP28VH1-like phosphatase (VHP)
02

Mechanism of action

Drugs targeting DUSP28 would theoretically act by inhibiting its phosphatase activity, thereby sensitizing cancer cells to apoptosis via reactivation of MAPK/ERK1/2 or other stress-responsive pathways

03

Biological functions

Dephosphorylation (removal of phosphate groups from proteins)Regulation of MAPK signaling pathways, including ERK1/2 and possibly p38Cell migrationApoptosisCell cycle regulationDrug resistance in cancer cells
04

Disease associations

Cancer (especially pancreatic and hepatocellular carcinoma)ChemoresistanceCell migration contributing to metastasis
05

Safety considerations

Specific safety concerns for targeting DUSP28 are not described in the available literature; potential challenges may include off-target effects due to similarity with other DUSPs and the physiological role of DUSP28 in normal tissue
06

Biomarkers

High DUSP28 expression in tumor tissue (especially in pancreatic and liver cancers) is associated with poor prognosis and chemoresistance; DUSP28 expression could be evaluated as a biomarker for drug sensitivity or metastatic potential
