Dual specificity phosphatase 4 (DUSP4) mRNA encodes the DUSP4 protein, also known as Mitogen-activated protein kinase phosphatase 2 (MKP-2), which is a critical regulator of the Mitogen-Activated Protein Kinase (MAPK) signaling pathway (UniProt P41230; NCBI Gene 1846). By dephosphorylating key kinases such as ERK, JNK, and p38, DUSP4 modulates cellular responses to growth factors and environmental stress, influencing cell proliferation, differentiation, and apoptosis (UniProt P41230; PMID: 22430633). In clinical oncology, DUSP4 mRNA levels serve as significant biomarkers; for instance, its loss is a known driver of resistance to MEK inhibitors and chemotherapy in triple-negative breast cancer (PMID: 22430633; PMID: 34552064). Conversely, its overexpression in other cancers, such as colorectal and pancreatic cancer, suggests a context-dependent role where it may promote tumor survival (PMID: 28655779; PMID: 20146334). Therapeutic strategies targeting DUSP4 mRNA primarily involve RNA interference (siRNA) or antisense oligonucleotides (ASOs) designed to restore or inhibit its expression to overcome drug resistance or halt tumor progression (PMID: 34552064; PMID: 32141356).
