Overview

Tri-specific tumor-anchoring target antigens refer to a therapeutic strategy in which a trispecific antibody or T-cell engager simultaneously binds to two different tumor-associated antigens (TAAs) to anchor the molecule to the cancer cell surface. This dual-targeting approach, often employing a logic-gated AND approach (e.g., targeting both BCMA and GPRC5D in multiple myeloma), is designed to enhance tumor selectivity and binding avidity while minimizing off-target effects on healthy tissues that express only one of the antigens. By requiring the presence of both antigens for optimal binding or activation, these molecules aim to reduce the risk of resistance caused by antigen escape, as the loss of a single antigen may not be sufficient for the tumor to evade the therapy. The third arm of the trispecific construct typically targets an immune effector receptor, such as CD3 on T cells or CD16A on NK cells, to facilitate the formation of an immune synapse and subsequent tumor cell lysis. This multi-antigen targeting approach represents a significant advancement in precision oncology, particularly for treating heterogeneous solid tumors and refractory hematologic malignancies. Prominent examples in clinical development include JNJ-79635322, which targets BCMA and GPRC5D on myeloma cells, and various experimental TriTCE platforms targeting HER2/HER3 or EGFR/EpCAM combinations.
