Overview

DUF3748 domain-containing protein, also known as YidR in bacteria, is a highly conserved protein found in various Enterobacteriaceae species, including Klebsiella pneumoniae and Escherichia coli [18, 21]. It plays a critical role in bacterial pathogenesis by mediating adhesion to host cells, invasion, and biofilm formation, making it a primary target for next-generation vaccines such as mRNA, DNA, and recombinant protein formulations [18, 25, 44]. In humans, the DUF3748 domain is present in the FAM210 protein family, specifically FAM210A and FAM210B, which are localized to the mitochondrial inner membrane [7, 11]. FAM210A is essential for mitochondrial translation and cardiac homeostasis, and its deficiency is linked to heart failure and sarcopenia [7, 27, 33]. FAM210B regulates heme biosynthesis and iron metabolism, and its expression levels serve as a predictive biomarker for response to mitochondrial-targeting cancer therapies like BTM-3566 [3, 30, 37]. Consequently, the DUF3748 domain represents a versatile target and biomarker across infectious disease, cardiology, and oncology.
