Overview

DXZ4 associated non-coding transcript 1, proximal (DANT1) is a long non-coding RNA transcribed toward the DXZ4 macrosatellite repeat on the human X chromosome from promoters flanking the array. DANT1, along with its counterpart DANT2, is developmentally regulated and shows changes in expression coincident with epigenetic alterations at the DXZ4 locus during cell differentiation. DANT1 is implicated in regulating heterochromatin formation at DXZ4 and may play a role in maintaining proper chromatin configuration, particularly in the context of X chromosome inactivation and embryonic stem cell pluripotency. Recent studies suggest that DANT1 may act as a competing endogenous RNA, influencing pluripotency by bridging regulatory networks specific to naive and primed states in human embryonic stem cells. Additionally, DANT1 has emerged as a potential circulating RNA biomarker for specific subtypes and disease status in childhood B-ALL, indicating a possible utility in disease stratification and risk assessment. No drugs are currently known to target DANT1, and it is not classified as a conventional therapeutic target.
