Overview

Dynactin-associated protein (DYNAP), also known as C18orf26 or DynAP, is a single-pass transmembrane protein that localizes to the Golgi apparatus and plasma membrane in a microtubule-dependent manner[2][4]. DYNAP interacts with the dynactin multiprotein complex, a critical regulator of dynein motor function and intracellular transport. DYNAP promotes oncogenic cell signaling, notably sustaining Akt kinase activation, which supports high rates of tumor cell proliferation and survival. Overexpression of DYNAP can drive transformation of NIH3T3 cells, and multiple isoforms generated by alternative splicing may modulate its activity. DYNAP is glycosylated at specific extracellular sites, with this glycosylation being necessary for its trafficking and transforming function[4]. Elevated DYNAP expression is predominantly observed in cancer cells, indicating a likely disease role in tumorigenesis and suggesting potential as a therapeutic target[2][4]. Ergosterol-derived compounds have been shown to selectively block DYNAP-mediated functions, supporting its promise as a cancer drug target[2].
