Overview

Dynactin subunit 1 (DCTN1) is the largest subunit of the dynactin macromolecular complex, a crucial component for dynein-mediated retrograde transport along microtubules[1][2][3]. The p150-glued isoform forms a flexible arm that mediates binding to microtubules, to the dynein intermediate chain, and to microtubule plus-tip tracking proteins[1][2]. Dynactin, together with cytoplasmic dynein, regulates processes including endoplasmic reticulum-to-Golgi transport, centripetal movement of lysosomes and endosomes, mitotic spindle formation, nuclear positioning, and axonogenesis[2][3]. DCTN1 mutations underlie several neurological diseases, such as amyotrophic lateral sclerosis, Perry syndrome, distal hereditary motor neuronopathy, and frontotemporal dementia[3]. DCTN1 is not currently a mainstream therapeutic target but plays a key role in normal neuronal cell functioning and neurodegeneration.
