Overview

Dynactin subunit 2 (DCTN2) is a 50-kDa component of the dynactin complex, which is essential for dynein-mediated, minus-end-directed transport along microtubules[1][2][3][5]. DCTN2 (also known as p50 dynamitin) serves as a core adapter, recruiting and anchoring dynein to diverse intracellular cargoes such as vesicles, organelles, and chromosomes. This action is crucial for processes including ER-to-Golgi transport, spindle formation, chromosome alignment, nuclear positioning, and neuronal development. Disruption of DCTN2, by overexpression or mutation, uncouples dynein from its cargo, leading to defects in intracellular trafficking, organelle localization, and cell division. Mutations in DCTN2 are associated with some neurodegenerative conditions, but there is no evidence that DCTN2 is a direct therapeutic target or interacts with drugs[1][2][3][4][5][6].
