Overview

Dynactin subunit 3 (DCTN3) is the smallest subunit of the dynactin complex, which is composed of 10 subunits ranging from 22 to 150 kDa. The dynactin complex is essential for the function of the motor protein cytoplasmic dynein, enabling ultra-processive transport of vesicles, organelles, and chromosomes along microtubules; DCTN3 activates dynein and is involved in many cellular processes including spindle formation, cytokinesis, chromosome movement, endosome trafficking, and axon development. DCTN3 is primarily an alpha-helical protein that binds directly to the largest dynactin subunit (p150) and exists only as part of the dynactin complex; it does not function as a stand-alone protein nor as a conventional drug target. Its primary clinical connection is in rare inherited forms of deafness; otherwise, it is not a known target for drug intervention nor used as a biomarker.
