Overview

Dynactin subunit 4 (DCTN4, also known as Dyn4 or p62) is a protein encoded by the DCTN4 gene in humans and is a structural subunit of the dynactin complex, which consists of multiple proteins. Dynactin is a key activator of cytoplasmic dynein, the minus-end directed microtubule motor, and is involved in a variety of cellular processes including organelle positioning, vesicle trafficking, and spindle formation during mitosis. The p62/DCTN4 subunit is located at the pointed end of the dynactin Arp1 rod, contributes to the stability of the complex, and is implicated in proper intracellular transport and cargo binding. Genetic variation in DCTN4 is associated with disease modification in cystic fibrosis and has been linked to non-syndromic deafness. In summary, DCTN4 is an essential, structural microtubule-associated protein within the dynactin complex but is not considered a direct receptor or enzyme target for therapeutics.
