Dynactin subunit 6 (DCTN6) is a protein that forms part of the dynactin complex, a key activator of the molecular motor cytoplasmic dynein, responsible for retrograde transport along microtubules and intracellular trafficking of organelles and protein cargoes. DCTN6 contains an RGD (Arg-Gly-Asp) motif in its N-terminal region, conferring adhesive properties and potential interactions with extracellular matrix proteins like fibronectin. In neurons, DCTN6 interacts with the endoplasmic reticulum and mediates retrograde transport; deficiency impairs ER dynamics and leads to dystrophic neurite formation, a feature linked to aging and Alzheimer’s disease. DCTN6 also regulates Golgi organization and is required for appropriate endo-lysosomal trafficking, partly by interacting with other membrane proteins. It serves as a binding partner for viral proteins (such as classical swine fever virus glycoprotein E2), supporting aspects of viral replication. DCTN6 mRNA expression carries prognostic significance in some cancers. The human protein is encoded by the DCTN6 gene on chromosome 8 and has high sequence conservation with mouse homologs, although its precise molecular roles are still being defined.
