Dynamin II (DNM2) is a large GTPase protein ubiquitously expressed throughout the body. It is essential for clathrin-mediated endocytosis, vesicle trafficking, and cytoskeletal organization. Mutations in DNM2 are associated with centronuclear myopathy, Charcot-Marie-Tooth disease, and acute lymphoblastic leukemia. Dynamin II functions by catalyzing the hydrolysis of GTP to provide energy for vesicle scission during endocytosis and filament remodeling.
Dynamins self‐assemble into helical polymers around the necks of budding vesicles. Upon GTP hydrolysis-driven conformational change they mediate membrane fission/scission events critical for intracellular trafficking processes such as endocytosis/exocytosis. This activity also underlies their role in remodeling other membranous structures within cells including those associated with the Golgi apparatus or during cytokinesis.
