Overview

Dynamin-related protein 1 (Drp1) is a large GTPase that plays a central role in mitochondrial fission. It belongs to the dynamin superfamily of proteins and is essential for cellular homeostasis, including mitophagy, mitochondrial distribution, ROS regulation, calcium signaling, and apoptosis. Drp1 dysfunction is implicated in various diseases, including neurodevelopmental disorders, neurodegenerative diseases, and cardiovascular disease.

Mechanism of action

Drp1 mediates mitochondrial division by assembling into multimeric structures that wrap around constriction points on mitochondria. Upon GTP hydrolysis, these structures constrict and sever the mitochondrial membrane.
