Overview

Dynein axonemal assembly factor 10 (DNAAF10), also known as WDR92 and Monad, is a highly conserved WD-repeat protein that acts as a key assembly/scaffolding factor specifically required for the stability of axonemal dynein heavy chains in the cytoplasm. DNAAF10 functions in the cytoplasmic pre-assembly of axonemal dynein arms, which are critical for the motility of cilia. It is thought to associate with a prefoldin-like cochaperone complex—helping maintain nascent dynein heavy chains in a state suitable for correct folding and preventing their aggregation. Loss or dysfunction of DNAAF10 impairs ciliary dynein arm assembly, leading to defects in ciliogenesis and cilia motility, as observed in model organisms and human ciliopathies such as primary ciliary dyskinesia. There are currently no drugs or known therapeutic agents specifically targeting DNAAF10, and there appear to be no established biomarker or safety considerations particular to this protein.
