Dynein axonemal heavy chain 17 (DNAH17) is a force-generating component of the outer dynein arms (ODAs) in the sperm flagellum, part of the axonemal dynein motor complex essential for microtubule-based motility. DNAH17 hydrolyzes ATP to produce force towards the minus ends of microtubules and plays a key role in sperm flagellar assembly and effective motility. Mutations in DNAH17 selectively disrupt sperm ODA function, resulting in isolated nonsyndromic male infertility (e.g., spermatogenic failure 39, asthenozoospermia, or multiple morphological abnormalities of the sperm flagellum), without affecting ciliary function in the respiratory tract[1][2][3]. DNAH17 is primarily relevant as a genetic cause and molecular marker for these infertility syndromes, not as a direct therapeutic target[1][3].
