Overview

Dynein axonemal heavy chain 3 (DNAH3) is a member of the axonemal dynein family, encoding a large microtubule-associated motor protein critical for the movement of motile cilia and flagella[1][2][3][4]. It is involved in generating the force required for ciliary and flagellar beating via ATP hydrolysis, playing an essential role in processes such as sperm motility and respiratory ciliary function[2][4]. DNAH3 is predominantly expressed in the testis, especially in the developing and mature sperm cells, where it is indispensable for proper flagellar assembly and function[3][4][5]. Loss-of-function or biallelic mutations in DNAH3 cause morphological and functional defects in sperm flagella, leading to male infertility phenotypes such as oligoasthenoteratozoospermia, characterized by low sperm count, poor motility, and abnormal morphology[4][5]. Variants in DNAH3 are also implicated in certain cancers, likely through effects on the cytoskeleton and cell motility. There are currently no known drugs that directly target DNAH3, and it is not a typical therapeutic target or drug receptor in clinical practice.
