Overview

Dynein axonemal light chain 1 (DNAL1) is an integral component of the outer dynein arms in cilia and flagella, serving as part of the molecular motor machinery responsible for ATP-dependent movement. It binds tightly to the dynein gamma heavy chain and interacts with alpha-tubulin, forming a crucial link required for effective ciliary action. DNAL1 is highly expressed in motile ciliated tissues such as respiratory epithelium and sperm flagella. Mutations in DNAL1 disrupt ciliary movement and cause recessive primary ciliary dyskinesia, while its involvement in HIV-1 replication suggests pleiotropic cellular roles. No drugs are known to directly target DNAL1, and therapeutic modulation is limited by its essential cellular function
