Dynein cytoplasmic 1 light intermediate chain 2 (DYNC1LI2) is a non-catalytic accessory subunit of the cytoplasmic dynein 1 motor protein complex which is essential for retrograde transport of vesicles and organelles along microtubules. DYNC1LI2 binds to dynein cargos and adaptor proteins, determining cargo specificity and regulating membrane trafficking events, lysosome and endosome positioning, and Golgi integrity. It plays a critical role in mitosis—especially spindle orientation and checkpoint control. Downregulation or mutation of DYNC1LI2 impairs vesicular trafficking and contributes to diseases such as cystinosis and certain ciliopathies. Post-translational modifications, such as arginine methylation, regulate its interaction with adaptors and diverse cargo, enabling the dynein complex to fulfill various specialized roles in the cell. Currently, it is not considered a direct therapeutic target and has no known drug interactions.
